Effects of Nimotuzumab Combined with γ-ray Radiation on Growth of the Esophageal Carcinoma Cells |

Abstract Objective To study the effect of inhibition of Nimotuzumab(h-R3) and ionizing radiation on the growth of human esophageal carcinoma cells. Methods Human esophageal carcinoma cells were treated with h-R3 or irradiation and the both, respectively. The cell growth inhibition was assessed by MTT. Cell cycle and apoptosis were analyzed by FACS. Results The growth of Ec109 cells treated with h-R3、radiation and h-R3 combined with irradiation were all inhibited. The inhibition rate was concentration-dependent and time-dependent. The group of h-R3 combined with irradiation resulted in greater inhibiting rate(43.74%), compared with h-R3 group and radiation group(P < 0.05). FACS revealed that combined treatment with h-R3 and radiation could not only increase cell cycle(G2 phases) arrest but also cell apoptosis of Ec109 cells. Cells of G2(31.67%) and apoptosis rate(23.62%) for h-R3 combined with radiation group were the highest. Conclusion h-R3 significantly inhibits the proliferation of Ec109 cells in vitro. Combined treatment with h-R3 and radiation shows synergism for the inhibition of esophagus carcinoma cells growth.
