Abstract Objective To investigate the effects of Rad51 depletion on sensitization to γ-radiation in osteosarcoma cell lines.Methods Four human osteosarcoma cells were examined for basal levels of Rad51 by western blotting analysis and RT-PCR.The suppression of Rad51 expression was performed using vector-based shRNA in HOS cells.Sensitivity of HOS cells treated with shRNA vector to γ-radiation was determined with colony formation assay and MTT assay.The apoptosis was determined by Flow cytometry.Results Rad51 showed higher basal levels in osteosarcoma cell lines than those in CCC-ESF-1 both in protein and RNA level.The suppression of Rad51 expression sensitized apoptosis of the cancer cells induced by γ-radiation.Conclusion Rad51 may serve as a promising therapeutic target for osteosarcoma.
