Abstract Objective To construct recombinant Rb94 gene adenovirus expression vector and to study its effect wirh γ-radiation in inhibiting the growth of breast cancer.Methods Total RNA was extracted from human embryo and reversed transcript to cDNA.Rb94 gene fragment was amplified by PCR and linked to pENTRTM 1A vector.Rb94 gene was recombinated with pAd/CMV/V5-DESTTM by LR recombinant and identified.Combined with γ-radiation, if produces an inhibiting effect on breast cancer cells.Results The recombinant Rb94 gene adenovirus expression vector was constructed successfully.The combined effect is better in inhibiting the malignant cells.Conclusion The recombinant Rb94 gene adenovirus expression vector can be used to investigate the function of Rb94 gene.Combination effect is stronger in inhibiting the growth of human malignant tumor cells.
