Abstract Objective To study the changes in main component of brain protein induced by internal contamination of enriched uranium in different level.Methods With low-temperature and high-speed centrifugation, ion exchange chromatography and dialysis, we separated and purified brain homogenate and got β-NGF from SD rat's brain.Then we tested the extracted β-NGF by neurite outgrowth test of PC12 cells and confirmed that it has high biological activity.Results Experiments showed that enriched uranium may lead to the reduction of the obtainable rate of β-NGF.It should be noted, that the reduction has a linear relation with enriched uranium dose.Meanwhile we found that the action of enriched uranium may lead to cleavage of brain protein, thereby enlarge the range of molecular weight spectrum of main component and increase the quantity of protein band in SDS-PAGE.Conclusion The radiotoxicological effects induced by enriched uranium were the cleavage of brain protein as well as the reduction of the obtainable rate of β-NGF.
